產(chǎn)品簡介: Caspase-10熒光檢測試劑盒提供了一種簡單方便的方法檢測能識別AEVD序列的 caspases活性.本檢測技術(shù)基于對底物AEVD -AFC（AFC：7-氨基-4-三氟甲基香豆素）.AEVD -AFC發(fā)生藍(lán)光（λmax = 400 nm）；caspase-10或者相關(guān)caspases酶切底物后，游離的AFC發(fā)出黃綠熒光（lmax = 505 nm），這種熒光可以被熒光計或者熒光微量滴定板讀數(shù)器定量分析.

Activation of ICE-family proteases/caspases initiates apoptosis in mammalian cells. The Caspase-10 Fluorometric Assay Kit provides a simple and convenient means for assaying the activity of caspases that recognize the sequence AEVD. The assay is based on detection of cleavage of substrate AEVD-AFC (AFC: 7-amino-4-trifluoromethyl coumarin). AEVD-AFC emits blue light (lmax = 400 nm); upon cleavage of the substrate by
caspase-10 or related caspases, free AFC emits a yellow-green fluorescence (lmax = 505 nm), which can be quantified using a fluorometer or a fluorecence microtiter plate reader.Comparison of the fluorescence of AFC from an apoptotic sample with an uninduced control allows determination of the fold increase in AEVD-dependent caspase activity.如與英文說明書有異，以英文說明書為準(zhǔn)。
