試劑盒簡介: Caspase熒光底物系列Plus包含即時(shí)可用的caspase-1/-2/-3/-5/-6/-8/-9 AFC 標(biāo)記的熒光底物和所有進(jìn)行caspase檢測(cè)實(shí)驗(yàn)的緩沖液.
產(chǎn)品背景: Caspase屬于半胱氨酸蛋白酶，相當(dāng)于線蟲中的ced-3，這些蛋白酶是引起細(xì)胞凋亡的關(guān)鍵酶，一旦被信號(hào)途徑激活，能將細(xì)胞內(nèi)的蛋白質(zhì)降解，使細(xì)胞不可逆的走向死亡.它們均有以下特點(diǎn)：①酶活性依賴于半胱氨酸殘基的親核性；②總是在天冬氨酸之后切斷底物，所以命名為caspase（cysteine aspartate-specific protease），稱之為凋亡酶；③都是由兩大、兩小亞基組成的異四聚體，大、小亞基由同一基因編碼，前體被切割后產(chǎn)生兩個(gè)活性亞基.
產(chǎn)品特點(diǎn): 所有Caspase比色底物都以即時(shí)可用的液體形式提供，使用方便，種類齊全.
1. Induce apoptosis in cells by desired method. Concurrently incubate a control culture without induction.
2. Count cells and pellet 1-5 x 106 cells or use 50-200μg cell lysates if protein concentration has been measured.
3. Resuspend cells in 50 μl of chilled Cell Lysis Buffer.
4. Incubate cells on ice for 10 minutes.
5. Add 50 μl of 2X Reaction Buffer and 1 μl DTT to each sample.
6. Add 5 μl of the 1 mM AFC conjugated substrates (50 μM final conc.) into each tube individually and incubate at 37°C for 1-2 hour.
7. Read samples in a fluorometer equipped with a 400-nm excitation filter and 505-nm emission filter. For a plate-reading set-up, transfer the samples to a 96-well plate. You may perform the entire assay directly in a 96-well plate.Fold-increase in caspase activity can be determined by comparing these results with the level of the uninduced control.如與英文說明書有異，以英文說明書為準(zhǔn)。
